This book describes a global assessment of stem cell engineering research, achieved through site visits by a panel of experts to leading institutes, followed by dedicated workshops. The assessment made clear that engineers and the engineering approach with its quantitative, system-based thinking can contribute much to the progress of stem cell research and development. The increased need for complex computational models and new, innovative technologies, such as high-throughput screening techniques, organ-on-a-chip models and in vitro tumor models require an increasing involvement of engineers and physical scientists. Additionally, this book will show that although the US is still in a leadership position in stem cell engineering, Asian countries such as Japan, China and Korea, as well as European countries like the UK, Germany, Sweden and the Netherlands are rapidly expanding their investments in the field. Strategic partnerships between countries could lead to major advances of the field and scalable expansion and differentiation of stem cells. This study was funded by the National Science Foundation (NSF), the National Institutes of Health (NIH) and the National Institute of Standards and Technology (NIST).

While the potential of stem cells is recognized, their proliferation and differentiation must be more precisely controlled to maximize the production of therapeutically relevant cells and for cell replacement therapies to minimize contamination with residual cells that can give rise to side effects. How can engineers make contributions to address these challenges? With contributions from pioneers and experts, Stem Cell Engineering: Principles and Practices highlights recent advances in the understanding of the cellular and molecular composition of the stem cell niche, as well as approaches to build upon this basic information to direct stem cell differentiation into therapeutically valuable lineages. The growing recognition of stem cells as an important and exciting field will continue to draw investigators with diverse backgrounds—from biology, engineering, and the physical sciences—and thereby enable further progress in these and other new directions. This book discusses advances made during the last decade that have led to increasingly defined culture systems for growing stem cells, starting from co-culture with feeder cells in the presence of serum to growth on synthetic substrates in defined medium. In addition to highlighting many recent advances, it underscores the need for future work.

Biology and Engineering of Stem Cell Niches covers a wide spectrum of research and current knowledge on embryonic and adult stem cell niches, focusing on the understanding of stem cell niche molecules and signaling mechanisms, including cell-cell/cell-matrix interactions. The book comprehensively reviews factors regulating stem cell behavior and the corresponding approaches for understanding the subsequent effect of providing the proper matrix molecules, mechanical cues, and/or chemical cues. It encompasses a variety of tools and techniques for developing biomaterials-based methods to model synthetic stem cell niches in vivo, or to enhance and direct stem cell fate in vitro. A final section of the book discusses stem cell niche bioengineering strategies and current advances in each tissue type. Includes the importance of Cell-Cell and Cell Matrix Interactions in each specific tissue and system Authored and edited by authorities in this emerging and multidisciplinary field Includes valuable links to 5-10 minute YouTube© author videos that describe main points

MSC (mesenchymal stem cells) have been reported to initiate revascularization after injury, to facilitate engraftment of blood-forming stem cells, and to reduce the incidence of graft-vs. host disease through their immune-suppressive qualities. Finally, bone marrow-derived MSC have been reported to home to areas of solid tumor revascularization, and thus may be used as delivery vehicles to target ablative agents into dividing tumor cells. Recently the characteristics of human MSC from adipose (fat) tissue have also been identified. The possibility of repairing tissues, speeding stem cell engraftment, and targeting solid tumors for specific killing, using MSC easily harvested from bone marrow, or better yet, from unwanted fat tissue, holds broad appeal, and is an intriguing possibility that could have dramatic effect on health care. This book has information on how to isolate, grow, and characterize MSC from marrow and fat, and gives important insight into how these cells may be used for gene delivery and cellular therapies in the future. Updates on emerging clinical trials are given.

I am very pleased to present this volume on engineering stem cells in Advances in Biochemical Engineering and Biotechnology. This volume stays abreast of recent developments in stem cell biology and the high expectations concerning the dev- opment of stem cell based regenerative therapies. Regenerative medicine is the focus of current biomedical research, with unique challenges related to scientific, technical and ethical issues of stem cell research, and the potential added value of connecting biomedicine with enabling techno- gies such as materials sciences, mechanical- and nano-engineering. Research activities in regenerative medicine include strategies in endogenous regeneration of injured or degenerated tissues by means of gene therapy or cell transplantation, as well as complex approaches to replace or reconstruct lost or malformed tissue structures, by applying tissue engineering approaches. In most cases, the speci- ized functional cell types of interest cannot be isolated from the diseased organ or expanded to a sufficient degree, and various stem and progenitor cell types rep- sent the only applicable cell source. In almost all cases, stem cells have to be engineered, sometimes for functional improvement, in many cases to produce large numbers of cells, and frequently to achieve efficient and specific differentiation in the cell type(s) of interest.
